  • 简介:InvitrodetectionmethodforthesensitivityofMagnaporthegriseatotricyclazolewasstudied,andthepotentialresistanceriskofblastdiseasetotricyclazolewasassessed.BothEC50ofhyphalmelanization(EC50-H)andminimuminhibitiveconcentrationofmelanizationinappressorial(MIC-A)byinhibitortricyclazoleshowedpositivecorrelationtotheEC50oftricyclazoleagainstblastdiseasetestedinvivo,withrelativeco-efficiency(R2)of0.8995and0.8244,respectively.However,stabilityandreproducibilityofEC50-HwerebetterthanthoseofMIC-A,suggestingthatitcouldbeusedtodetectthesensitivityofM.griseatotricyclazoleinvitro.TricyclazolesensitivityoftheprogeniesderivedfromsinglesporesofthemostsensitiveisolateDY2andtheleastsensitiveisolateGY6detectedinsensitivitymonitoringin2000wasnotstable,withmeanEC50valuesof4.4968μg/mLand5.4010μg/mL,respectively,indicatingthatthedifferenceinEC50betweenDY2andGY6wasnotcausedprobablybyresistancevariation.EC50ofGY6didnotincreasesignificantlywhencontinuouslyselectedfortwentygenerationsundertheselectionpressureoftricyclazoleinvivo.However,thesensitivityofDY2wasdecreasedby10-foldafterselectedfortwentygenerations.TheresultssuggestedthattricyclazolewasstilllowresistanceriskforM.griseainChina.

  • 简介:Genotypingbysequencing(GBS)istherecentapproachofnext-generationsequencingtechniquefordiscoveringandgenotypingsinglenucleotidepolymorphisms(SNPs)incropspecies.Genotypicvariationstudies(SNPsandinsertion-deletions/InDels)wereperformedusingfourricelinesbasedonGBSdatabyaligningtothereferencegenomeNipponbare.LocalaromaticricelandraceTulaipanjiwascrossedwithRanjit,andtwodistinctlineswereidentifiedfromtheprogenies:onelinewithawnsandaromatraitsandtheotherwithoutawnsandaroma.TotalnumberofSNPsandInDelsidentifiedwere52810and4327atreaddepth10,respectively.OutofthetotalpolymorphicSNPs/InDels,16490wereintergeneric,7812wereinsidegene,and4435wereintronic.Phylogenetically,Tulaipanjiwasclosertothereferencegenomenipponbare.Basedonrecurrentparentgenomeanalysis,outof10013alleles,92.52%wasintrogressedintoprogeny-awnfromTulaipanjiand7.48%fromRanjit,whereasprogeny-awnlesscarried89.19%allelesfromRanjitandonly10.81%allelesfromTulaipanji.Inaddition,progeny-awnwasthehighestheterozygous(83.88%)andprogeny-awnlesswastheleast(2.24%)atthisfifthgenerationofrecombinantinbredlines.TheseSNPvariationsmaybelinkedtothephenotypictraitsandcanbeutilizedincropimprovementthroughlinkagemapping.TheseresultssuggestthataddingahighdensityofSNPmarkerstoamappingorbreedingpopulationthroughGBShasagreatvaluefornumerousapplicationsinricebreedingandgeneticsresearch.

  • 简介:ThaijasminericeKDML105isconsumedaroundtheworld.BKOS,PKOSandTKOSarenewcultivarsproducedfromlow-energyionbeaminductioninKDML105.ThepurposeofthisstudyistocomparethemorphologyandanatomybetweenKDML105andthethreenewcultivars.Seedsofthefourcultivarsweregerminatedandgrowninpotsuntilfloweringphase.Theplants'organswereobservedandthelengthsofculms,ligules,leavesandpaniclesweremeasured.Leafsurfaceareawascalculatedandnumbersofroots,spikeletsandtillerswerecounted.BKOSandPKOShadsignificantlyshorterculmsthanKDML105andTKOS.ThelargestleafareawasfoundinKDML105followedbyTKOS,BKOSandPKOS,respectively.NumbersofrootsandtillersinBKOSandTKOSweresignificantlyfewerthanthoseinKDML105andPKOS.ThenumberofspikeletsperplantinBKOSwasthelowestamongallcultivars.Foranatomicalcomparison,crosssectionsofculmsandrootswereobserved.Allplantshadasimilararrangementoftissues,butthenumberandsizeofcellsweredifferent.Furthermore,longitudinalsectionsofculmsshowedthatthelengthsofepidermalandparenchymacellsweredirectlyrelatedwiththelengthoftheculm.Tocomparetheleaves,bothstomataandepidermalcellswerecountedandthelengthsoftheguardcellsweremeasured.ThelengthsofguardcellsofBKOSandPKOSwereshorter,butthestomataldensityandthestomatalindexweresignificantlygreaterthanthoseofKDML105.ForTKOS,thoughthelengthofguardcellswasshorterthanthatinKDML105,thedifferencewasnotsignificant.However,thestomataldensityandstomatalindexweresignificantlyhigherthanthoseinKDML105.
