Involvement of regulatory volume decrease in the migration of nasopharyngeal carcinoma cells

ThetranswellchambermigrationassayandCCDdigitalcameraimagingtechniqueswereusedtoinvestigatetherelationshipbetweenregulatoryvolumedecrease(RVD)andcellmigrationinnasopharyngealcarcinomacells(CNE-2Zcells).Bothmigratedandnon-migratedCNE-2Zcells,whenswollenby47%hypotonicsolution,exhibitedRVDwhichwasinhibitedbyextracellularapplicationofchloridechannelblockersadenosine5'-triphosphate(ATP),5-nitro-2-(3-phenylpropylamino)benzoicacid(NPPB)andtamoxifen.However,RVDrateinmigratedCNE-2Zcellswasbiggerthanthatofnon-migratedcellsandthesensitivityofmigratedcellstoNPPBandtamoxifenwashigherthanthatofnonmigratedcells.ATP,NPPBandtamoxifenalsoinhibitedmigrationofCNE-2Zcells.TheinhibitionofmigrationwaspositivelycorrelatedtotheblockageofRVD,withacorrelationcoefficient(r)=0.99,suggestingafunctionalrelationshipbetweenRVDandcellmigration.WeconcludethatRVDisinvolvedincellmigrationandRVDmayplayanimportantroleinmigratoryprocessinCNE-2Zcells.
