  • 简介:Theexperimentwascarriedouttoassessthereactionofdifferentcategoriesofricegenotypesviz.,resistant,susceptible,hybrid,scented,popularandwildinresponsetotheinfestationbyriceleaffolder(RLF),Cnaphalocrocismedinalis(Guenee)andtoexplorethepossibleuseofthesegenotypesindevelopingRLF-resistantricevarieties.Thechangesofvariousbiochemicalconstituentssuchasleafsolubleprotein,phenol,ortho-dihydroxyphenol,tanninandenzymesviz.,peroxidase,phenylalanineammonialyase(PAL)wereassessedspectrophotometricallyinallthericegenotypesbeforeandafterRLFinfestation.Theproteinprofilewasanalyzedusingsodiumdodecylsulphate-polyacrylamidegelelectrophoresis(SDS-PAGE)method.Asignificantconstituentofbiochemicalcontentsuchastannin,phenolandortho-dihydroxyphenolhasbeenincreasedalongwithenzymeactivitiesofperoxidaseandPALintheinfestedresistant(Ptb33,TKM6andLFR831311)andwildricegenotypes(OryzaminutaandO.rhizomatis).Adecreaseinleafproteincontentwasevidentinvariablyinalltheinfestedricegenotypes.Itisalsoevidentthatthecontentsofbiochemicalssuchasphenol,orthodihydroxyphenolandtanninwerenegativelycorrelatedwithleaffolderdamage.However,leafproteincontentwaspositivelycorrelatedwiththedamagebyriceleaffolder.SDS-PAGEanalysisfortotalproteinprofilingofhealthyandC.medinalis-infestedgenotypesrevealedtheenhancedexpressionofahighmolecularweight(>97kDa)proteininallthegenotypes.Besides,therewasalsoanincreasedinductionofa38kDaproteininC.medinalisinfestedresistantgenotypes,whichwasabsentinuninfestedplants.ThepresentinvestigationprovedthattheelevatedlevelsofbiochemicalsandenzymesmayplayavitalroleinriceplantsresistancetoRLF.

  • 简介:Plantassociatedbacteriaarepromisingalternativestochemicalfertilizersforplantgrowthandyieldimprovementinaneco-friendlymanner.Inthisstudy,riceassociatedbacteriawereisolatedandassessedformineralphosphatesolubilizationandindole-3-aceticacid(IAA)productionactivityinvitro.Sixpromisingstrains,whichweretentativelyidentifiedasphylotaxonPseudochrobactrumsp.(BRRh-1),Burkholderiasp.(BRRh-2),Burkholderiasp.(BRRh-3),Burkholderiasp.(BRRh-4),Pseudomonasaeruginosa(BRRh-5andBRRh-6)basedontheir16SrRNAgenephylogeny,exhibitedsignificantphosphatesolubilizingactivityinNationalBotanicalResearchInstitutephosphategrowthmedium,andBRRh-4displayedthehighestphosphatesolubilizingactivity,followedbyBRRh-5.ThepHoftheculturebrothdeclined,resultinginincreaseofgrowthrateofbacteriaatpH7,whichmightbeduetoorganicacidsecretionbythestrains.InpresenceofL-tryptophan,fiveisolatessynthesizedIAAandthemaximumIAAwasproducedbyBRRh-2,followedbyBRRh-1.ApplicationoftwomostefficientphosphatesolubilizingisolatesBRRh-4andBRRh-5byrootdipping(colonization)ofseedlingandsprayingatthefloweringstagesignificantlyenhancedthegrowthandgrainyieldofricevarietyBRRIdhan-29.Interestingly,applicationofbothstrainswith50%ofrecommendednitrogen,phosphorusandpotassiumfertilizersproducedequivalentorhighergrainyieldofricecomparedtothecontrolgrownwithfullrecommendedfertilizerdoses,whichsuggeststhatthesestrainsmayhavethepotentialtobeusedasbioinoculantsforsustainablericeproduction.

  • 简介:Agreenhousepotexperimentwasconductedwithhybridrice(OryzaSativaL.)inordertostudyNStatusandutilizationintherhizosphereofrice,Theexperimentwascomposedofthreetreatments:withoutN,15NH4-Nand15NO3-N.Plantrootswereseparatedfromthesoilbyanyloncloth,and1mmincrementsofsoil,movinglaterallyawayfromtheroots,weretakenandanalyzedforvariousNfroms.ThelabelledNintheplantsrangedfrom67\51%to69.24%ofthetotalamountofNabsorbedbythericeseedlingswiththelabelledfertilizerNtreatments.ThisshowsthattheNinthePlantscamemainlyfromthefertilizers.However,theNabsorbedbythericeseedlingsaccountedforlessthan35%ofthetotalamountoftheNdepletioninthesoilnearthericeroots,indicatinganimportantNlossintherhizosphereofrice.Thesoilredoxpotential(alltreatments)andtheconcentrationofthelabelledNO3-N(thelabelledNH4-Ntreatmentonly)decreasedasthedistancefromthericerootsincreasedintherhizosphereofrice.Incontrast,theconcentrationofthelabelledNH4-Nincreasedasthedistanceuincreasedinthesamesoilzone.Theseresultssuggestedthatnifrificationoccurredinthesoilaroundthericeroots.Therefore,thereasonfortheNlossintherhizosphereofricemightbetheNO3movementintothereductivenon-rhizospheresoil(submerged)wheredenitrificationcantakeplace.

  • 简介:Itisnecessarytounderstandthebacterialpopulationsassociatedwithricesoastoprovidemoreinformationandnaturalresourcesforeffectivemanagementofmajordiseasesinrice.Asurveyonscreeningandidentificationofgram-positivebacteriawasconductedduring1998-2004.Sevenhundredandfifty-sixricesampleswerecollectedfromZhejiang,Jiangsu,FujianandYunnanProvinces,China.Over1000bacterialisolateswereisolatedandtestedforcolonymorphology,pathogenicity,andsomecharacteristicsofbacteriologyincludingGramstaining,fluorescentpigmentonKingsmediumBandmicroscopicobservationforendospore.Togetherwithfivestandardreferencestrains,74representativegram-positivebacterialisolateswereconfirmedbyBiologandgaschromatographicanalysisoffattyacidmethylesters.FivebacterialspeciesofBacillusandotherthreegenerawereidentifiedandisolatesfromBacillussublitisandBacillusmegaterium,exhibitedthemosteffectiveinhibitionagainstthepathogensofsheathblightandbakanaediseaseofrice.AfewisolatesfromBacilluspumilusandBacillusmegateriumshowedweakvirulentonricetogetherwithsomevirulentisolates,dskshouldbeconsideredwhenisolatesfromthesespecieswerescreenedforbiocontrolagents.
