摘要 Inordertocreateanewmimicofglutathioneperoxidase(GPx),bioimprintingwasusedtogenerategluta-thione(GSH)bindingsiteandchemicalmodificationwasusedtoincorporatecatalyticgroupselenocystine(Sec).Humanserumalbumin(HSA)andS-substituteddinitrophenylglutathione(GSH-S-DNP)werechosenastheimprintedmatrixandimprintingtemplate,respectively,togenerateaGSH-imprintedprotein(GSH-HSA)bybioimprinting.SecwasincorporatedintotheGSH-HSAbychemicalmodificationtogiveanewGPxmimic(Se-GSH-HSA).Se-GSH-HSAdisplayedconsiderablyhigherGPxactivitythannon-printedHSA(Se-HSA).TheenzymicpropertiesandkineticsofSe-GSH-HSAwerestudied.Moreover,Se-GSH-HSAwasconfirmedtohavestrongerantioxidantabilitytoprotectmitochondriaagainstoxidativedamagewithferroussulfate/ascorbate-inducedmitochondriadamagemodel,indicatingthatSe-GSH-HSAhaspotentialapplicationinmedicine.
