摘要 Chip-basedflow-throughPCRimplementsthePCRasacontinuousprocessfornucleicacidanalytics.Thesampleistransportedinawindingchannelthroughtemperaturezonesrequiredfordenaturation,annealingandextension.Mainfieldsofapplicationarethemonitoringofcontinuousprocessesforrapididentificationofcontaminantsandqualitycontrolaswellashighthroughputscreeningofcellsormicroorganisms.Amodulararrangementwithfiveheatingzonesforflow-throughPCRisdiscussedandevaluated.Thespecialheaterarrangementallowstheimplementationofupto40cyclesonthefootprintofamicroscopeslide,whichisplacedontopofa5zonesheatingplate.Liquid/liquidtwophaseflowofPCRreactionmixtureandmineraloilhavebeenappliedtocreateasegmentedflowprocessscheme.Inthatway,thedevelopedsystemmayprovideflow-throughPCRasaunitoperationforthedropletbasedmicrofluidicsplatform.ThesingleuseofdisposabledevicesiscommonlypreferredduetothesensitivityofthePCRprocesstocontaminations.All-glassmicrofluidicchipsanddisposablechipdevices,madefrompolycarbonateasareplicationwithidenticallygeometry,havebeenfabricatedandtested.Forthefirsttime,microchannelgeometrieswithnearlycircularprofiledevelopedbyall-glasstechnologyhavebeentransferredtomassfabricationbyinjectioncompressionmolding.Bothdeviceshavebeensuccessfullyappliedforthedetectionofthetumorsuppressorgenep53.Althoughproductyieldandselectivityoftheamplificationprocessdonotdependonthechipmaterial,awelldefined,reliablesegmentedflowregimecouldonlyberealizedintheall-glasschip.
