摘要 Proteomicsisthestudyofproteinsandtheirinteractionsinacell.WiththecompletionoftheHumanGenomeProject,theemphasisisshiftingtotheproteincomplimentofthehumanorganism.Becauseproteomereflectsmoreaccuratelyonthedynamicstateofacell,tissue,ororganism,muchisexpectedfromproteomicstoyieldbetterdiseasemarkersfordiagnosisandtherapymonitoring.Theadventofproteomicstechnologiesforglobaldetectionandquantitationofproteinscreatesnewopportunitiesandchallengesforthoseseekingtogaingreaterunderstandingofdiseases.High-throughputproteomicstechnologiescombiningwithadvancedbioinformaticsareextensivelyusedtoidentifymolecularsignaturesofdiseasesbasedonproteinpathwaysandsignalingcascades.Massspectrometryplaysavitalroleinproteomicsandhasbecomeanindispensabletoolformolecularandcellularbiology.Whilethepotentialisgreat,manychallengesandissuesremaintobesolved,suchasmininglowabundantproteinsandintegrationofproteomicswithgenomicsandmetabolomicsdata.Nevertheless,proteomicsisthefoundationforconstructingandextractingusefulknowledgetobiomedicalresearch.Inthisreview,asnapshotofcontemporaryissuesinproteomicstechnologiesisdiscussed.
